Freeze-fracture electronmicroscopy of developing optic nerve axons in rat demonstrated clusters of intramembranous particles on the external face in some fibers before myelination. These particle aggregates may represent precursors of nodes of Ranvier. The ability of the membranes of CNS fibers to develop regional specializations, which do not depend on the presence of either myelin or mature axoglial junctions, may be significant in terms of normal ontogenesis and membrane reorganization after demyelination.
